Types of APIs: Small Molecules, Peptides and Biologics
Last updated: September 2026. For formulators and procurement teams comparing the three classes of active pharmaceutical ingredients before specifying a source.
Small molecules are chemically synthesised, stable, and the cheapest of the three to scale. Peptides sit in between: short amino-acid chains, precise in action, fragile to make, usually injected. Biologics are large molecules grown in living cells, costly and tightly regulated. Most oral generics still run on small molecules, and that is the class our API portfolio is built on.
Small Molecule APIs
A small molecule API is a chemically synthesised compound, usually under about 900 daltons. Its structure is fully defined, verifiable by IR and HPLC, and reproducible batch after batch. That is why small molecules still carry most of the world’s oral dose forms: analgesics like paracetamol and ibuprofen, the penicillin antibiotics, statins such as atorvastatin, and the SGLT2 inhibitor APIs used in type 2 diabetes.
They also scale well. The route is fixed, intermediates are isolable, the equipment is standard reactor trains. Scale-up is a heat-transfer problem, not a question of keeping a cell line alive. Impurities are controlled against ICH Q3A, residual solvents against ICH Q3C, and the API manufacturing process follows the same logic at 5 kg and at 500 kg.
Peptide APIs
Peptides are short chains of amino acids. Because they mimic sequences the body already recognises, they reach targets a small molecule cannot hit cleanly. Insulin is the obvious example; the GLP-1 agonists used in diabetes and obesity are the current one, and oncology is the other main use, where a peptide binds a receptor expressed on tumour cells.
Making them is harder. Every residue added is another coupling step and another chance of a deletion sequence. Peptides react to temperature, pH and oxidation, so cold chain and protective packaging become part of the specification rather than a precaution. Gut proteases break most of them down before absorption, which is why the route is normally injection.
Biologic APIs
Biologics are large molecules made by living systems: monoclonal antibodies, vaccines, gene therapy vectors. A cell line is engineered to express the protein, grown in a bioreactor, then harvested and purified. They changed the standard of care in several cancers and in rheumatoid arthritis, and opened treatment routes in rare inherited disorders that had none.
The cost is complexity. With a biologic the product is defined by the process: change the cell line, the feed or the purification train and you change the molecule. Regulators respond with comparability studies and a longer approval path. Our overview of API regulations and standards covers the GMP framework underneath all three classes.
Comparing the Three
| Small molecules | Peptides | Biologics | |
|---|---|---|---|
| Size | Under about 900 Da | About 500-5,000 Da | Tens to hundreds of kDa |
| Made by | Chemical synthesis | Solid-phase or recombinant synthesis | Living cells in bioreactors |
| Structure | Fully defined | Sequence-dependent | Process-dependent |
| Typical route | Oral | Injection | Injection or infusion |
| Stability | High | Moderate | Low, cold chain standard |
| Relative cost | Lowest | Middle | Highest |
Small molecules for volume and affordability. Peptides where a chronic indication needs precision. Biologics where nothing smaller works. No single class covers a full therapeutic range, which is why buyers watching API manufacturing trends in India usually hold suppliers across more than one.
Where Bio-Synth Fits
We are Biochemical and Synthetic Products Private Limited, and we have manufactured APIs at Balanagar, Hyderabad since 1943. The work is small-molecule chemistry: antipsychotics, anticoagulants, antihistamines, anti-tuberculosis agents, the SGLT2 class. The published range sits on our API portfolio page.
Our site holds ISO 9001:2015 certification from DNV Business Assurance UK Limited under UKAS accreditation, certificate 156792-2014-AQ-IND-UKAS, first issued 20 August 2002, scoped to “Development, manufacture and sale of active pharmaceutical ingredients (APIs), drug intermediates, biological products & liver injectable”. A WHO-GMP certificate from the Drugs Control Administration, Government of Telangana (L.Dis.No. 150458/TS/2024, valid to 5 June 2027). Manufacturing follows ICH Q7 principles. Details sit on our quality management system page.
A certificate covers what it names. Facility-level certification says the quality system was audited, not that every product on site is individually certified. Ask which API appears on which certificate, and read the annexure.
