Ethopabate API Role in Poultry Coccidiosis Treatment

Ethopabate API Role in Poultry Coccidiosis Treatment

Last updated: August 2026 | Scope: how Ethopabate works against Eimeria, why it is paired with amprolium, and what that means for feed formulators sourcing the API. For identity, grades and general veterinary use, see the companion post.

Coccidiosis in poultry is caused by Eimeria parasites that damage the gut lining and cut growth. Ethopabate is a folate-pathway antagonist that blocks the parasite’s ability to make the folic acid it needs to multiply. It targets the intestinal species, especially E. acervulina, but not E. tenella, so it is used in combination with amprolium. Bio-Synth supplies BP (Vet) and USP grade Ethopabate for these combination premixes.

Coccidiosis in poultry: the problem

Coccidiosis is a protozoal infection of the intestinal tract. Chickens pick up Eimeria oocysts from litter, the parasite invades the gut wall, and each species settles into its own section of the intestine. E. acervulina favours the upper small intestine, E. maxima the mid-gut, E. brunetti the lower intestine, and E. tenella the caeca.

The gut lining is destroyed as the parasite multiplies, nutrient absorption drops, and in severe cases birds bleed and die. Even subclinical infection costs feed efficiency and weight gain. In dense housing, oocysts spread fast, which is why prevention through feed, rather than treating sick birds, has been the standard approach for decades.

Anticoccidial drugs in poultry fall into two broad groups: ionophores, which are fermentation products, and synthetic chemicals. Ethopabate belongs to the second group.

How Ethopabate works

Ethopabate API is a structural analogue of para-aminobenzoic acid (PABA). Eimeria cannot absorb pre-formed folate from the host; it has to synthesise its own, and PABA is the starting material. Ethopabate competes with PABA in that pathway, so the parasite is starved of the folate it needs for nucleic acid synthesis and cannot complete its replication cycle.

That is also why the drug is safe for the bird. Chickens obtain folate from feed and do not run the same synthetic pathway, so the block is selective.

Timing matters. Ethopabate shows its peak activity around the fourth day of the parasite’s life cycle, during the asexual multiplication stages in the gut wall. It suppresses the population before the damaging later stages build up, rather than killing established parasites.

For readers who want the molecule’s identity and physical properties, the companion post on what Ethopabate API is covers the CAS number, formula, solubility and grades.

Spectrum: what it covers and what it does not

Ethopabate has a defined and fairly narrow spectrum:

  • Good activity: E. acervulina, the most widespread intestinal species in broilers.
  • Partial activity: some strains of E. maxima and E. brunetti.
  • No activity: E. tenella, the caecal species responsible for bloody caecal coccidiosis.

That last gap rules Ethopabate out as a stand-alone product. It has only ever been used in combination.

The amprolium–Ethopabate combination

Amprolium is a thiamine (vitamin B1) antagonist. It blocks thiamine uptake by the parasite and is strongest against E. tenella, precisely the species Ethopabate misses. Put the two together, and the intestinal and caecal species are both covered by two different mechanisms.

Historically, the combination was introduced with Ethopabate at 4 ppm in feed and later raised to 40 ppm, always alongside amprolium. Two independent modes of action also make resistance slower to develop than it would be against either agent alone, which is one reason this pairing has stayed in use for so long.

For the formulator, this means Ethopabate is rarely purchased as a single-active premix. The sourcing question is whether the Ethopabate API blends cleanly with amprolium and the carrier at a consistent, low inclusion rate. Particle size, bulk density, and flow are the properties that decide that.

Where it fits in a control programme

Producers rotate or shuttle anticoccidials to slow resistance. A typical shuttle uses one chemical in the starter feed and switches to an ionophore in the grower, or the other way round. Amprolium-Ethopabate combinations are one of the chemical options in that rotation.

Ethopabate is a synthetic antiprotozoal, not an antibacterial, so it sits outside the antibiotic growth-promoter debate. Regulatory status and withdrawal periods still vary by market and are set at the finished-product level, so formulators should verify local rules before use.

What premix manufacturers should check when sourcing

A coccidiostat premix is only as reliable as the API going into it. Four things to verify:

  1. Monograph compliance. BP (Vet) or USP, with a Certificate of Analysis showing assay of 96.0–101.0% on the dried basis and the identification and melting-range tests passed.
  2. Impurity control. ICH Q3A/Q3B principles for related substances and ICH Q3C or VICH GL18 for residual solvents.
  3. Manufacturing site credentials. Bio-Synth produces Ethopabate at its Hyderabad facility, which holds a WHO-GMP certificate from the Drugs Control Administration, Government of Telangana, and is run in line with ICH Q7 principles. The quality page describes the QC and documentation system.
  4. Batch-to-batch consistency. At 40 ppm in feed, a small assay drift in the API becomes a meaningful dose error in the bird. Ask for CoAs from several batches, not one.

How an API like this is made, from starting materials through purification and drying, is covered in the general API manufacturing process overview.

FAQ

Does Ethopabate treat coccidiosis or prevent it?

Both, but its main value is prophylaxis. Fed continuously, it suppresses the parasite before clinical disease develops.

Why is Ethopabate not effective against E. tenella?

Its folate-pathway action works well against the intestinal species but not the caecal species. Amprolium covers E. tenella.

Is Ethopabate an antibiotic?

No. It is a synthetic antiprotozoal, not an antibacterial.

What inclusion rate is used?

The combination was introduced at 4 ppm of Ethopabate and later 40 ppm, always with amprolium. Finished-product rates are set by the formulator and local regulator.

What grades does Bio-Synth offer?

BP (Vet) and USP, each with a Certificate of Analysis. Reach the team through the contact page for a specification sheet.
